Methods 1. Quick's Method.-The method in use in this laboratory has been basically that of Quick, StanleyBrown, and Bancroft (1935), as modified later by Quick himself and others (Biggs and Macfarlane, 1949). In this method a fresh saline emulsion is prepared daily from a stock of acetone-dried human brain powder, which is kept stored in a vacuum desiccator at 40 C. 2. Toohey's Method.-This method obviates the necessity of using dried brain extracts (Toohey, 1941, 1950; Owen and Toohey, 1941). Fresh human brain stripped of membranes and blood vessels is simply macerated with 0.5% w/v phenol in 0.85% w/v saline. The emulsion is thoroughly shaken and kept in a tightly stoppered container at 40 C. In

Protein Thrombokinase and Lipoid Thromboplastin as Distinct Factors with Complementary Functions *

Two classes of biologic agents can accelerate the production of thrombin from partially purified prothrombin. One class is well represented by platelets, the other by a factor obtainable from the plasma globulins. Representatives of both classes are found not only in blood, but also in crude tissue extracts. These two classes of agents are distinct; and they serve different functions in the clo...

Studies on the differences in activity of lung and brain thromboplastin.

Recent reports have suggested that thromboplastin (Tpl) prepared from lung may reflect more accurately than does brain Tpl the changes produced in the prothrombin complex by coumarin derivatives (1, 2). As evidence for this, it has been observed that as factor VII concentration decreases in the plasma of Dicumarol '-treated patients, the prothrombin time obtained with lung Tpl becomes dispropor...

Comparison of the thrombotest with the modified Quick test. Drug-induced hypoprothrombinemia countered with phytonadione.

COUMARIN and indanedione derivatives are widely used anticoagulants. These compounds induce hypoprothrombinemia by depressing the synthesis of factor II (prothrombin), factor VII (proconvertin), factor IX (plasma thromboplastin component, Christmas factor), and factor X (StuartPrower factor), and this hypoprothrombinemia is neutralized most effectivelv by phytonadione (vitamin K1). The laborato...

Prothrombin Time in Dicoumarol Therapy.

In the course of controlling dicoumarol therapy given (a) as a postoperative routine to prevent thrombosis (fifty-five cases) and (b) therapeutically in cases of established venous thrombosis (nine cases), certain peculiarities have been observed in the behaviour of the prothrombin time, particularly in relation to dicoumarol haemorrhage. Two sources of thromboplastin have been used, Russell vi...
